t is now well accepted that defined architectural compartments within the cell nucleus can regulate the transcriptional activity of chromosomal domains within their vicinity. However, it is generally unclear how these compartments are formed. The nuclear periphery has received a great deal of attention as a repressive compartment that is implicated in many cellular functions during development and disease. The inner nuclear membrane, the nuclear lamina, and associated proteins compose the nuclear periphery and together they interact with proximal chromatin creating (...) a repressive environment. A new study by Harr et al. identifies specific protein–DNA interactions and epigenetic states necessary to re‐position chromatin to the nuclear periphery in a cell‐type specific manner. Here, we review concepts in gene positioning within the nucleus and current accepted models of dynamic gene repositioning within the nucleus during differentiation. We report the identification of a recurrent, 520-kb 16p12.1 microdeletion associated with childhood developmental delay. The microdeletion was detected in 20 of 11,873 cases compared with 2 of 8,540 controls and replicated in a second series of 22 of 9,254 cases compared with 6 of 6,299 controls. Most deletions were inherited, with carrier parents likely to manifest neuropsychiatric phenotypes compared to non-carrier parents. Probands were more likely to carry an additional large copy-number variant when compared to matched controls. The clinical (...) features of individuals with two mutations were distinct from and/or more severe than those of individuals carrying only the co-occurring mutation. Our data support a two-hit model in which the 16p12.1 microdeletion both predisposes to neuropsychiatric phenotypes as a single event and exacerbates neurodevelopmental phenotypes in association with other large deletions or duplications. Analysis of other microdeletions with variable expressivity indicates that this two-hit model might be more generally applicable to neuropsychiatric disease. © 2010 Nature America, Inc. It is widely recognized that ‘global’ symmetries, such as the boost invariance of classical mechanics and special relativity, can give rise to direct empirical counterparts such as the Galileo-ship phenomenon. However, conventional wisdom holds that ‘local’ symmetries, such as the diffeomorphism invariance of general relativity and the gauge invariance of classical electromagnetism, have no such direct empirical counterparts. We argue against this conventional wisdom. We develop a framework for analysing the relationship between Galileo-ship empirical phenomena on the one hand, and (...) physical theories that model such phenomena on the other, that renders the relationship between theoretical and empirical symmetries transparent, and from which it follows that both global and local symmetries can give rise to Galileo-ship phenomena. Consisting of fewer than two hundred verses written in an obscure if not impenetrable language and style, Patanjali's Yoga Sutra is today extolled by the yoga establishment as a perennial classic and guide to yoga practice. As David Gordon White demonstrates in this groundbreaking study, both of these assumptions are incorrect. Entire moral philosophies have been rejected for ruling out the possibility of failure. This “fallibility constraint” (also sometimes called the “error constraint”) cannot be justified by appealing either to Wittgensteinian considerations about rules or to the moral importance of alternate possibilities. I propose instead that support for such a constraint in ethics can be found in the Strawsonian reactive attitudes. I then use the constraint to reveal hidden weaknesses in contemporary contstitutivist strategies to ground moral normativity such as Christine Korsgaard’s, (...) and also to reveal hidden strengths in historical accounts of morality such as Bishop Butler’s. We will have reason to reject any moral theory that makes constitutivism’s mistake, but only because we have reason not to reject the fallibility constraint itself. The way this ethical fallibility can be justified suggests a general principle that could be used to justify fallibility constraints in other normative domains such as practical reason, epistemology, the philosophy of language, and the philosophy of mind. Over the past fifty years, a number of approaches to the recovery of the multiple pasts of Hinduism have held the field. These include that of the discipline of History of Religions as it is constituted in North America as well as those of the Hindu nationalists, the col and post-colonial historians, and the Subaltern Studies School. None of these approaches have proven satisfactory because, for methodological or ideological reasons, none have adequately addressed human agency or historical change in their (...) accounts of the pasts out of which modern-day Hinduism has emerged. The Hindu nationalist historians hark back to an extended Vedic golden age in which religious practice remained unchanged until the corruptions spawned by the Turkish invasions of the eleventh century. Many Western indologists and historians of religion specializing in Hinduism never leave the unalterable ideal worlds of the scriptures they interpret to investigate the changing real-world contexts out of which those texts emerged. The colonial and postcolonial historians focus on the past two hundred years as the period in which all of the categories through which India continues to interpret itself—including Hinduism—were imposed upon it from without. Adducing examples of Hindu practitioners and thinkers from the colonial period, subaltern theorists and others argue that historical thought is itself alien to the authentic Indian mind. This article suggests a number of interpretive strategies for retrieving the multiple Hinduisms of the past and of the medieval period in particular as that time out of which most modern-day practices of Hinduism emerged. These include an increased emphasis on non-scriptural sources and a focus on regional traditions. (shrink)

As David White explains in the Introduction to Tantra in Practice, Tantra is an Asian body of beliefs and practices that seeks to channel the divine energy that grounds the universe, in creative and liberating ways. The subsequent chapters reflect the wide geographical and temporal scope of Tantra by examining thirty-six texts from China, India, Japan, Nepal, and Tibet, ranging from the seventh century to the present day, and representing the full range of Tantric experience--Buddhist, Hindu, Jain, and even (...) Islamic. Each text has been chosen and translated, often for the first time, by an international expert in the field who also provides detailed background material. Students of Asian religions and general readers alike will find the book rich and informative. The book includes plays, transcribed interviews, poetry, parodies, inscriptions, instructional texts, scriptures, philosophical conjectures, dreams, and astronomical speculations, each text illustrating one of the diverse traditions and practices of Tantra. Thus, the nineteenth-century Indian Buddhist Garland of Gems, a series of songs, warns against the illusion of appearance by referring to bees, yogurt, and the fire of Malaya Mountain; while fourteenth-century Chinese Buddhist manuscripts detail how to prosper through the Seven Stars of the Northern Dipper by burning incense, making offerings to scriptures, and chanting incantations. In a transcribed conversation, a modern Hindu priest in Bengal candidly explains how he serves the black Goddess Kali and feeds temple skulls lentils, wine, or rice; a seventeenth-century Nepalese Hindu praise-poem hammered into the golden doors to the temple of the Goddess Taleju lists a king's faults and begs her forgiveness and grace. An introduction accompanies each text, identifying its period and genre, discussing the history and influence of the work, and identifying points of particular interest or difficulty. The first book to bring together texts from the entire range of Tantric phenomena, Tantra in Practice continues the Princeton Readings in Religions series. Five medieval Sanskrit-language descriptions of a fabulous technique for extracting mercury from the “wells” in which it naturally resides are shown to be remarkably similar to accounts preserved in Chinese and Syriac. Whereas the Sanskrit and Chinese versions date from no earlier than the thirteenth century C.E., the Syriac version dates from no later than the tenth century. The present article first compares and contrasts these three alchemical narratives, and then suggests that all three are perhaps related to a broader (...) and far more ancient Indo-European mythic tradition of a deity associated with the phenomenon of “Fire in Water,” as attested in Vedic, Avestan, Roman, Irish, and Greek sources. All eight of these witnesses appear to attest to ancient religious and scientific traditions relative to geothermal phenomena. (shrink)

Our understanding and praxis of spiritual formation is only as sound as our theology of the Holy Spirit. Unfortunately, there is a common shortcoming that leaves our theology of the Spirit less than fully capable of rendering an adequate account of the Holy Spirit's agency in transforming a person into the likeness of Christ. Theology of the Holy Spirit commonly emphasizes victorious and genteel themes while failing to articulate adequately that the Spirit is a direct agent in transformative suffering. Some (...) of the Comforter's most important sanctifying work is exceedingly discomforting. This article focuses on these less palatable aspects of the Spirit's work in an attempt to tell a fuller story about the Spirit. By adding often-neglected dimensions and textures to our theology of the third Person, we are more capable of articulating the full range of the Spirit's role in formation. (shrink)

Public acceptance is often seen as a key reason why water-recycling technology is rejected. A common assumption is that projects fail because the general public is unable to comprehend specialist information about risk and the belief that if the public were better informed, they would accept change more readily. This article suggests that rhetoric about acceptance is counterproductive in progressing sustainability as it does not address issues relating to institutional arrangements and reinforces a dichotomy between expert and lay groups. Instead, (...) it is argued that institutional change is needed to build opportunities for constructive public engagement. The failure to implement sustainable water use through recycling can be understood as the result of several factors including present cost structures for water, institutional conservatism, administrative fragmentation, and inadequate involvement of communities in planning. Achieving sustainable water use through recycling may require better coordination between agencies and integrated government policies. (shrink)

_A powerful treatise that demonstrates the existence of altruism in nature, with surprising implications for human society_ Does altruism exist? Or is human nature entirely selfish? In this eloquent and accessible book, famed biologist David Sloan Wilson provides new answers to this age-old question based on the latest developments in evolutionary science. From an evolutionary viewpoint, Wilson argues, altruism is inextricably linked to the functional organization of groups. “Groups that work” undeniably exist in nature and human society, although special (...) conditions are required for their evolution. Humans are one of the most groupish species on earth, in some ways comparable to social insect colonies and multi-cellular organisms. The case that altruism evolves in all social species is surprisingly simple to make. Yet the implications for human society are far from obvious. Some of the most venerable criteria for defining altruism aren’t worth caring much about, any more than we care much whether we are paid by cash or check. Altruism defined in terms of thoughts and feelings is notably absent from religion, even though altruism defined in terms of action is notably present. The economic case for selfishness can be decisively rejected. The quality of everyday life depends critically on people who overtly care about the welfare of others. Yet, like any other adaptation, altruism can have pathological manifestations. Wilson concludes by showing how a social theory that goes beyond altruism by focusing on group function can help to improve the human condition. (shrink)

Perfectionism is an underexplored tradition, perhaps because of doubts about the grounds, content, and implications of perfectionist ideals. Aristotle, J.S. Mill, and T.H. Green are normative perfectionists, grounding perfectionist ideals in a normative conception of human nature involving personality or agency. This essay explores the prospects of normative perfectionism by examining Kant’s criticisms of the perfectionist tradition. First, Kant claims that the perfectionist can generate only hypothetical, not categorical, imperatives. But insofar as the normative perfectionist appeals to the normative category (...) of personality or agency, rather than a biological category of humanity, it can represent perfectionist demands as categorical imperatives. Second, Kant accepts a moral asymmetry in which we aim at our own perfection but at the happiness, rather than the perfection, of others. However, the importance of autonomy in normative perfection explains why the perfectionist should recognize a self/other asymmetry. Indeed, when we see how the normative perfectionist can answer Kant’s criticisms while respecting Kant’s own claims about the connection between rational nature and moral requirements, we can see the basis for a normative perfectionist interpretation of Kant’s own ethical theory. Insofar as Kantians and normative perfectionists both base ethical demands on an appeal to rational nature, they face a common worry that the appeal to rational nature is empty or incomplete. Normative perfectionists have more and less concessive responses to this worry, providing perfectionist explanations of various apparently non-perfectionist goods. Even if we end up being pluralists about the good, perfectionist elements play an important role. Finally, because the normative perfectionist, like the Kantian, grounds its ideals and requirements in a conception of persons as rational agents, it provides a promising account of the rational authority of perfectionist demands. This comparison of normative perfectionist and Kantian essentials gives us reason to take the normative perfectionist tradition seriously. (shrink)

In this paper, we delve into the significant impact of recent advancements in Artificial Intelligence (AI) on the future landscape of work. We discuss the looming possibility of mass unemployment triggered by AI and the societal repercussions of this transition. Despite the challenges this shift presents, we argue that it also unveils opportunities to mitigate social inequalities, combat global poverty, and empower individuals to follow their passions. Amidst this discussion, we also touch upon the existential question of the purpose of (...) human life in a world where machines predominantly perform work. Additionally, we explore the contentious issue of the moral status and rights of AI entities, contemplating the scenario where machines evolve to become sapient and sentient. The paper concludes with a call to action, emphasizing the necessity to acknowledge these potential outcomes and proactively seek solutions, as part of an inevitable journey through the evolution of human society shared with intelligent machines. In their book, Equal Is Unfair, Watkins and Brook argue that equality of income and wealth is not needed in order to engage in the creative work required for human flourishing. One can live a successful life even though others have more resources and opportunities. It is contended here that this argument is convincing, but contrary to Watkins and Brook, it does not suffice to rule out all justifications for redistribution.

Addressing fake news requires a multidisciplinary effort The rise of fake news highlights the erosion of long-standing institutional bulwarks against misinformation in the internet age. Concern over the problem is global. However, much remains unknown regarding the vulnerabilities of individuals, institutions, and society to manipulations by malicious actors. A new system of safeguards is needed. Below, we discuss extant social and computer science research regarding belief in fake news and the mechanisms by which it spreads. Fake news has a long (...) history, but we focus on unanswered scientific questions raised by the proliferation of its most recent, politically oriented incarnation. C'est que, au premier abord, sa philosophie - et celle construite avec Félix Guattari - se présentent comme une sorte d'encyclopédie de mouvements aberrants. C'est ce qu'il a cherché dans le cinéma, dans la peinture, la littérature, les sciences, dans l'histoire des sociétés, dans la philosophie, et partout. Ainsi par exemple, les déformations des figures de Francis Bacon, les non-sens de Lewis Carroll, les lignes destructrices et créatrices des populations nomades à (...) travers l'histoire, la démence du système capitaliste, les processus schizophréniques qui forment l'inconscient, toutes les forces qui traversent la vie et la pensée. Voilà ce qu'il faut d'abord expliquer : la présence de ces mouvements aberrants Mais on ne peut pas s'en tenir là. La philosophie ne peut pas se contenter de les décrire ou d'en dresser la carte, elle doit rendre raison de ces mouvements. D'où un deuxième aspect, tout aussi important : il faut produire les logiques irrationnelles de ces mouvements aberrants. C'est l'une des grandes nouveautés de l'oeuvre de Deleuze et Guattari : créer de nouvelles logiques, loin des modèles rationnels classiques, mais aussi loin de modèles contemporains, comme le marxisme ou le structuralisme orthodoxes des années 1960-1980. Mais ces logiques n'ont rien d'abstrait, au contraire : ce que veulent faire Deleuze et Guattari, c'est dégager les logiques de tous les modes de peuplements de la terre. Par peuplement, il ne faut pas seulement entendre les populations humaines, mais les populations géologiques, animales, cosmiques ou encore les populations mathématiques, esthétiques, philosophiques qui peuplent la pensée des hommes. Quelle est la logique de tous ces peuplements? Poser une telle question, ce n'est pas seulement demander des explications, mais exiger des justifications. Par exemple, pour le capitalisme : de quel droit se déploie-t-il sur la terre? De quel droit s'approprie-t-il nos cerveaux pour le peupler d'images et de sons? De quel droit asservit-il les corps? Aux logiques que le capitalisme met en oeuvre, ne faut-il pas opposer d'autres logiques, celles justement des mouvements aberrants? Dès lors, la philosophie devient inséparable de combats contre toutes les formes d'organisation - politique, sociale, philosophique, esthétique, scientifique - qui tentent de nier, de conjurer, d'écraser l'existence de ces mouvements aberrants.
